One post-doctoral position is available in the Laboratory of Molecular Structure Characterization, Institute of Microbiology (IMIC), Videnska 1083, CZ 142 20 Prague 4, Czech Republic. The duration of this post-doctoral fellowship is 12 months in the first stage with possible extension to the second year. Start of the fellowship is upon mutual agreement; we prefer to receive your application by March 31, 2018. Monthly fellowship will be 1150 Eur in gross, upon signing tax exemption form the income represents 870 EUR netto/month. Accommodation in campus hostel is available (monthly rent 190 EUR). The position linked to the project entitled “Clinical ion mobility spectrometry” consists of two individual subsections, i.e. (1) Intraoperative mass spectrometry in burn surgery and (2) The diagnostics of infectious diseases by metabolomics approaches. Both parts are carried out under the umbrella of the National Sustainability Program of the IMIC funded by the Czech Ministry of Education, Youth and Sports of the Czech Republic (LO1509). The development, construction and clinical application of a tangential electrosurgical knife compatible with for online characterization of human tissues by rapid evaporation ionization mass spectrometry is planned for the burn surgery. Necrotic and potentially vital tissue types will be quantitatively distinguished. This work will further be supported by FTICR MSI combined with metabolomics on nano-HPLC-IM-QTOF. Both tools will also be employed in development of non-invasive diagnostic approaches useful for infections caused by BSL-2 pathogens in man or animal models. The applicant must hold a PhD degree within one of the following disciplines – analytical chemistry, biology, molecular biology, immunology, microbiology, pharmacology or biochemistry. Applicants who have submitted their doctoral thesis can also apply if the defence is expected before Apr 30, 2018.
Our laboratory combines three analytical tools for the characterization of molecular structure: mass spectrometry, nuclear magnetic resonance spectroscopy and electron microscopy. The instrumentation is spread in buildings A, C, U and L on campus. In addition to own scientific research, the contractual work is provided for academic and industrial partners (reQuest application). Mass spectrometry group is focused mainly to proteomics, peptidomics and lipidomics fields. For biomolecular separation prior to mass spectrometry 1D/2D chromatographic or gel methods are being employed. The collisionally induced dissociation and electron transfer dissociation are available for de novo sequencing. For molecular 2D visualization MALDI, nano- DESI and NALDI imaging mass spectrometry protocols have been developed and implemented on our 12T FTICR or ion mobility spectrometers. Structural biology approaches provided to the community are represented by chemical crosslinking, hydrogen-deuterium exchange and molecular modeling. These services are provided by our partner Laboratory of Structural Biology based in Biotechnology and Biomedicine Center of the Academy of Sciences and Charles University in Vestec.
Nuclear magnetic resonance group operates a four channel 700 MHz instrument with a cryo-probe dedicated to protein analysis (2013). An older (2009) 400 MHz instrument has been dedicated to analysis of low molecular weight natural compounds, their semisynthetic derivatives or biotransformation products. The third, four channel 600 MHz NMR instrument running in HPLC-NMR configuration, is used for metabolomics projects. It also can be on-line linked to a Q-TOF mass spectrometer (micrOTOF-Q III).
Microscopy group runs multiple electron as well as optical microscopy equipment. In late 2014 a new scanning electron microscope FEI NanoSEM 450 was installed. It is equipped with CBS detector for high resolution imaging of non-conductive samples and EDAX Octane Plus detector for EDS analysis and elemental mapping. In accord with other laboratory groups extensive service measurements are being carried out for the whole biological campus in Krc as documented by considerable number of collaborations and joint publications. The effective instrument sharing with Palacky University in Olomouc, Charles University in Prague, Purkinje University in Ústí nad Labem and the Institute of Chemical Technology in Prague is enabled through our part or full time scientists.
